III. Video on Factors Affecting Enzyme Function
A quick 10-minute video on factors affecting enzyme function
(the guy has an annoying voice, but if you can stand it, it’s helpful.)
IV. Important Notes on Simulation Lab
Password: 784G8
Constant Values:
Enzyme Concentration is constant throughout at 1 x 10-6
Temperature should kept constant at human body temperature: 37 °C
Exception: If you have Amylase in Thermophiles, set at 70 °C
Substrate concentration should be kept constant at the highest possible concentration: 0.01 M
pH should be kept constant at the optimal pH for your enzyme (determined by experiment I)
Variables
When manipulating temperature, start at 10 °C and increase by 10 °C each round
When manipulating substrate concentration, start at 0.001 M and increase by 0.001 M each round
Units
Concentration = Molar (M)
Temperature = °C
Rate of Reaction = mol/min
V. Review of Scientific Notation
This Program Uses Old Calculator Scientific Notation
1E-006 = 1 x 10-6
Numbers expressed in scientific notation are either super big (positive exponent) or super small (negative exponent)
We will be seeing a lot of super small numbers during the course of this experiment
4.2 x 10-8 M = 0.000000042 M
Subtract one from the absolute value of the exponent to figure out the number of zeros to put after the decimal point
Which is bigger?
1.12 x 10-12 or 1.12 x 10-4
9.97 x 10-6 or 1.17 x 10-6
What is 0.00045 in scientific notation?
The number of zeros after the decimal point is one less than the absolute value of the exponent
ANS: 4.5 x 10 ^ (-4)

Figure 1 The effect of the concentration of catalase (drops) on the rate of hydrogen peroxide decomposition (kPa/min). Pressure is collected with Vernier interface and pressure sensor attachment. This pressure data is analyzed using the “06B Enzyme (Pressure)” file within the LoggerPro software program. Temperature is kept constant at 21 °C.
V. Advice for Writing Your Enzyme Paper: Legends
A good legend will have:
Figure number
Description of Graph
Technology used
Constants
Example for Experiment 1: Changing [Catalase]
Note that for experiment 3 and 4 (manipulating Temp and pH), the [enzyme] and the [substrate] are constant. You will need to indicate that in the legend.
For legends describing simulated experiments, you will need to note that the EME Corporation: Enzyme Investigations program was used to generate data.
Dedicate three pages of your paper to the discussion.
Easiest way to organize it = 6 paragraphs
Background about enzymes: what are they, how do they work, what do they do (name, year). How do the four variables affect enzyme function (name, year). Information of assigned enzyme: where it functions, what it does, optimal pH, optimal temperature (name, year).
How increasing [enzyme] affects reaction rate according to the pre-existing scientific body of knowledge (name, year). Your hypothesis on how increasing [catalase] will affect reaction rate. Summarize results of Catalase experiment 1 and discuss why the data followed that pattern (Figure 1). Explain error. Transition to talking about assigned enzyme and the existing body of knowledge with regard to the effect of varying this particular enzyme’s concentration on reaction rate (name, year).
How increasing [substrate] affects reaction rate according to the pre-existing scientific body of knowledge (name, year). Your hypothesis on how increasing [H2O2] will affect reaction rate. Summarize results of Catalase experiment 2 and discuss why the data followed that pattern (Figure 2). Explain error. Transition to talking about assigned enzyme and the existing body of knowledge with regard to the effect of varying substrate concentration on reaction rate (name, year). Summarize results of Simulated Substrate Experiment with Assigned Enzyme, discussing why the data followed that pattern and if this is similar to (or different from) the results of the same experiment with Catalase (Figure 3).
How increasing temperature affects reaction rate according to the pre-existing scientific body of knowledge (name, year). Your hypothesis on how increasing temperature will affect reaction rate. Summarize results of Catalase experiment 3 and discuss why the data followed that pattern (Figure 4). Explain error. Transition to talking about assigned enzyme and the existing body of knowledge with regard to the effect of varying temperature on reaction rate (name, year). Summarize results of Simulated Temperature Experiment with Assigned Enzyme, discussing why the data followed that pattern and if this is similar to (or different from) the results of the same experiment with Catalase (Figure 5).
How increasing pH affects reaction rate according to the pre-existing scientific body of knowledge (name, year). Your hypothesis on how increasing pH will affect reaction rate. Summarize results of Catalase experiment 4 and discuss why the data followed that pattern (Figure 6). Explain error. Transition to talking about assigned enzyme and the existing body of knowledge with regard to the effect of varying pH on reaction rate (name, year). Summarize results of Simulated pH Experiment with Assigned Enzyme, discussing why the data followed that pattern and if this is similar to (or different from) the results of the same experiment with Catalase (Figure 7).
Wrap it up by a) mentioning the effect of manipulating each of the variables again and b) suggesting another variable that may have an effect on reaction rate to test in future experiments.
