Solving Your SUMMER Unknown Project
After receiving your unknown, please remember your assigned number. There will be no discussions in class. That means no talking, laughing, whispering, comparing, etc. DO NOT MAKE EYE CONTACT WITH YOUR PEERS. The only help you may use in class: a) your notes (the ones that belong to you, no sharing), b) your lab manual, c) Bergey’s manual.
Make note of everything you do.
1st: Complete a Streak Plate on: 1 PEA, 1 EMB, and 2 BHI plates, or 2 TSA plates, or 2 NA plates
2nd: Determine the shape of your unknown by completing a Simple Stain or Negative Stain. Note the results. Determine the gram reaction of your unknown by completing a KOH test. Note the results. Confirm/Determine the gram reaction and morphology of your unknown by completing a Gram Stain. Leave your Gram-stained slide focused under oil immersion . Note the results.
3rd: Report your gram reaction and morphology.
I will confirm the gram reaction and morphology by viewing your slide under the microscope. (If
your slide is not focused and/or not in oil immersion, I will deduct points.) After confirmation,
you will receive your assigned chart. Use the chart to determine your unknown.
In addition to the Gram Stain, KOH, and morphology checkpoints, other checkpoints MAY be announced throughout the process to make sure you are headed in the right direction. The chart you receive will guide you throughout your journey. Once you have completed your journey and determined your unknown genus and species, you must turn in a report. The ELECTRONIC report is due two days after you determine the unknown and no later than Tuesday, July 1st at 4:00 p.m. After that date and time, I WILL NOT ACCEPT YOUR UNKNOWN REPORT. I REPEAT, I WILL NOT ACCEPT YOUR UNKNOWN REPORT.
Please upload all reports directly to Canvas. If you have any trouble uploading your report into Canvas, you may email the report to me before the deadline.
Your report should be written using the following guidelines:
· Cover Page: your name, class time/days, unknown #, and unknown name (genus and species)
· Introduction: Summarize the details of your genus only (history, medical importance, interesting facts, etc.). No more than two paragraphs (400 words) please. No less than SEVEN sentences.
· Test/Experiments: name, purpose, and results of EVERY TEST you perform (including EMB, PEA, Gram Stain, KOH, and Simple/Negative)
· Conclusion: Summarize your genus and species (history, habitat, etc.). No more than two paragraphs (400 words) please. No less than SEVEN sentences.
· Reference Page: include at least three references (at least one book)

I may not call roll during the “unknown days”, but it is your responsibility to be here and on time. If no one shows up after 15 minutes, I will close up the lab and you will NOT be able to complete any experiment(s) or record any results that day. You may not come in on any other lab time (not mine or any other class). Use your time wisely.
Those concerned with point deductions, they are as follows (numbers in parentheses are for each offense):
• If you ask for equipment that is already out. Look before you ask. ( -5)
• If you tell me silly things such as, “…the citrate test is gram negative.” ( -8)
• Forgetting your unknown number ( -5), not using PPE ( -7), or walking away from your lit Bunsen burner ( -20).
• If I catch you talking, whispering, or helping someone else ( -5 to -10). If you need me to answer a basic question such as, “What dye is used for a negative stain?” ( - 10)
• If I find your organism under the microscope after you have stated that nothing is on your slide ( -15).
• If you do not discard your materials by the last day ( -3). *for each tube and for each plate
• Not including all the information I asked for in your unknown report ( varies per item).
• And anything else I deem as cause for a deduction ( ??)
