You have fluorescein labeled anti-CD4 and phycoerythrin labeled anti-CD8 antibodies for the analysis of mouse cells by flow cytometry. You use these antibodies to stain thymocytes and lymph node cells from 2 mice strains: normal mice and RAG-1 gene knock-out mice. You already know that in peripheral blood of normal mice, 66% of the T cells are CD4pos T cells and 33% are CD8pos T cells. You also know that lymph nodes of normal mice contain a few cells that are not T cells (i.e., NK cells, macrophages, dendritic cells, and B cells). Based on the labelled flow cytometry FACS plots below, (i) complete this worksheet table to indicate which quadrants (A, B, C, and/or D) you would expect to see cell populations in this experiment and (ii) explain why your answer is correct. You can refer to the attached page 2, as needed. In your results, you should also consider cells in your sample that are not T cells, such as B-cells, NK cells, macrophages, and dendritic cells.
