Abstract

Bacterial endospores are highly resistant to temperature, radiation, desiccation and chemical agents which are oval or spherical structures. vegetative cell gives rise to the endospore. During procedure Malachite green was used as primary stain, safranin was used for counterstain and acid alcohol was used to decolorizing drive primary stain into the cell. As result endospores were present in bright green color whereas vegetative cells appeared brownish red or pink in color. The best image is found using the 40x and 100x compound microscope.

Introduction

To visualize bacterial endospore, the endospore could be used. The formation of endospores is made from some genera bacteria, for instance, Bacillus. When bacteria forms into spores, they can survive in a less habitat conditions. Also, spores are unique, resistant to heat, radiation and chemical. The purpose of endospore staining is to differentiate spore formers from non-spore formers and to differentiate bacterial spores from others vegetative cells. The spore is stained in a manner so that it can be distinguished from the vegetative part of the cell in which the staining technique is called endospore staining. Spores are structures which are typically resistant to heat, radiation, chemicals and other agents that are typically lethal to the organism. a spore forming, motile, rod-shaped, Gram-positive, facultative aerobe is called bacillus subtilis. With an optimal growth temperature from 25-35 degrees Celsius, it is mostly found in vegetation and soil. The subtilis can be used for study of endospore formation into a bacterium. (Aryal, 2020)

Method and materials

The procedure starts with cleaned isopropanol. The first process is to prepare a smear of bacillus subtilis, air dried and fixed it by heat. After that, we placed paper towel over the slide without the strip touching the edge of the slide. In addition, the slide was heated by placing it on the top of the boiling water. The slide was continuously stained with malachite green to prevent it from drying or evaporating and this process was done for five minutes. The slide than wash drained gently, and MG was gently rinsed of with our squeezing bottle. Over the staining tray slide with safranin was stained for one minute and then drained and washed with water. Finally, the slide left for to air dry.

Results and discussion

(40x microscope) (100xmicroscope)

When completing the staining procedure, the slides were gently blotted with lenses cleaning paper or Kim wipes before viewing under the microscope. Then each slide was examined using compound light microscope at 4x, 10x, 40x, and 100x magnifications, but best image is found when using 40x and

100x. Using 40x and 100x compound light microscope in Endospore staining endospores are present in sample, these will retain the malachite green stain so, it appear the bright green whereas vegetative cells are stained red by the safranin counter stain and appear brownish red or pink in color.
