Please read the article by Hirsch et al. (2017) and write a paper review. Please follow the guideline and the rubric below. I have uploaded a shorter version with marked paragraphs. (Hirsh_2027_ann). Use this version to follow the guideline. Regeneration of the entire human epidermis using transgenic stem cells https://www.nature.com/articles/nature24487 Study Guide: General Background: This paper describes a case of treating a boy suffering from a genetic disease JEB. The authors made transgenic stem cells using the boy's skin stem cells by adding the DNA to produce the deficient protein. They grew them in tissue culture and transplanted to the boy to replace his entire epidermis. I have highlighted some questions in each section. Please include the answer to these questions in your review. Abstract: Summary

• This section summarizes its findings. Note how the first two sentences describe the background, and the third sentence starts talking about their results. An abstract should mostly contain the findings or results, not an introduction. Your abstract should follow this, and the introduction should not exceed more than 30 % of the abstract.

The main article-Paragraphs of each section is labeled as P1, P2…. Introduction P1: This section describes JEB. Do not worry about the gene themselves. Just a general description of problems is important. (1) What is hemidesmosome? Search the internet and include a simple description.

P2: This section describes background: success and problem in the area, and the summary of the main result. (2) What are holoclone, meroclone, and paraclone? Search the internet and include a simple description. The patient: describes conditions of the patient Regeneration of epidermis by transgenic culture P1~P6: describes general experimental procedures and results The authors made the transgenic cell by adding the DNA which codes for the normal DNA. This DNA will randomly be inserted in an area in each cell and produce the normal protein. P7~P8: describes results in Fig. 2 Probes are used to visualize only certain molecules or organelles. Based on this figure, the authors claim that transgenic cells successfully renewed the epidermis. (3) Do you agree with this claim, and why or why not? Explain using Fig 2. Integration profile of transgenic epidermis-do not include this section This section describes how the cells now express the normal protein and why they think the DNA they used to express the normal protein is safe. The transgenic epidermis is sustained by holoclones P1: The patient received about 400 million dividing cells, and16 million holoclone cells among them. Per cm2 of the skin, 46000 dividing cells and 1800 holoclone cells were transplanted. P2: The authors describe two hypotheses.

1. All the dividing cells are equally good at dividing 2. Only a few long-lived stems cells provide all the cells of the epidermis

The DNA for the transplanted cells is randomly inserted (integrated) in the genome, and each cell's DNA location is different. By checking the location of DNA integration, you can figure out whether the cells are from the same cell or different cells. P3: Ignore this section P4: To verify either of the two hypotheses, they checked the locations of DNA in cells of PG (right after transplant), 4 (4M) months, and (8M) months later. (Fig. 4c). The blue color in the pie chart shows the percentage of cells having the same integration site as holoclones. (4) Does this show that only a small percentage of stem cells provide most of the cells? The rest of the content could be freestyle, but make sure to include the following content. Follow the guidelines provided.

• Begin with a summary of relevant background information and statement of the major question being addressed.

• A discussion of the experimental approaches- explaining the question/hypothesis, methodology, results, and interpretation of the data (1 page),

• a summary of the overall conclusions, including a discussion of whether hypotheses were supported or not. (1/2 page)

• A bibliography, not part of the 3-page limit, should be included. For the term paper, following the correct bibliography format is important. You can use any bibliography format typically found in a scientific journal. This includes "in-text" citation and detailed information in the "reference" section. Grading Rubric
