Wet Mount Slide Preparation

Definition:

A Drop of water or stain on slide with specimen, then view under a microscope.

Materials Needed

Glass Slide

Cover slip.???????

Water

Stain (methylin blue and why do we use it?

Wooden stick

Forceps

Dropper

Preparation

Procedure:

1.Add one drop of water on the slide.

2.Gently scratch your cheek from inside.

3.Mix it with the drop of water on the slide.

4.Gently place the cover slip on top of the sample..avoid air bubbles.

5.Place it under the microscope.

6.Repeat the procedure using methylene blue instead of water.

 

* Cheek cells

Procedure:

Add one drop of water on the slide.

Carefully, peel thin layer of the onion and place it on the slide that contain water.

place the cover slip gently and avoid air bubbles.

place it under microscope,,use the objectives 10 and 40 .

Repeat the procedure using methylin blue instead of water.
