For example so let’s say you’re looking at a particular protein Musashi 2 (MSI2) that is seen to be increased in people who are on a WW diet and you’re trying to see if that protein is linked to an increase to being better toleration of a particular chemo drug Paclitaxel that targets Triple negative Breast Cancer.
Aim1: Determine the expression level of MSI2 in triple negative breast cancer cell lines. in however many breast cancer lines you have and compare it to the normal control lines you have. Determine endogenous levels of MSI 2 in all breast cancer cell lines and compare to the control lines via western blot. . And then in your first aim you would also do several tests with cells that have that protein knocked out to see what happens to those cancer cells.
Then in aim 2 you would test the effect of that protein expression with the chemo drug in breast cancer cells and the also treat the chemo drug with the breast cancer cell lines knocked out to see if there is a difference. And you need to include the mechanism of action of the drug you’re using and the concentration or IC50 you want to use.
Then the third aim is using mice. So for example Like 20 mice who are on a WW diet and have breast cancer, 20 mice who are on WW and don’t Have breast cancer and a group of 20mice who have breast cancer but are not on a special diet. So you would look first at giving these mice their respective diets over a course of 15 days or presentation of tumor and And then do a western blot to see if the protein is there and at what level it is. Also treat the mice with the chemo drug and do imaging to see if you see the tumor size decrease or metastasis court or not. And so on so forth.
Your aims should connect but they also should be able to stand on their own just in case something doesn’t work out with one of the aims. So maybe tight pool at a series of proteins or whatever it is you want to look at and go from there.
