Prin ciple Lipoprotein lipase hydrolyses triglycerides to glycerol and free fa tty acids . The glycerol fo~~ ed with ATP in the presence of glycerol kinase form s glycerol 3 phosphate which is 1 ox1d1sed by the enzyme glyceroi phosphate oxidase to fo rm hydrogen peroxide. The hydrogen peroxide further re acts with phenolic compound and 4-aminoantipyrine by the catalytic action of peroxidase to fonn a red coioured qu i;,or. eir nir, e dye complex . Intensity of • the colou r form ed is directly proportional to the amount of triglycerides present in the sample.

Storage / stability
Contents are stable at 2-8°C ti ll the expiry mentioned on the labels .
Reagent Preparation Reagents are re ady to use . Working reagent : Pour the contents of 1 bottle of L2 (Enzyme Reagent 2) into 1 bottle of L 1 (Enzyme Reagent 1 ). This working reagent is sta ble for at least 8 weeks when stored at 2-8' C. Upon storage the working reagent may develop a slight pink colour however this does not affect the performance of th e reagent. Alternatively fo r fl exibility as much of work ing reagent may be made as and when desired by mixing together 4 parts al 11 (Enzyme Reagent 1) & 1Jart of L2 ( Enryme Reagen\2 ). Alternatively Q~ ml of L 1 and 0.2 ml of L2 may also be used instea d of 1 ml of the wor mg reagent directly during the assay.
