Read this paper by Bonafont et al (2019) , and answer the following questions that evaluate the methods and purpose of the study:

1. CRISPR/Cas-9 was used to edit a specific region of DNA, what was the purpose of editing that specific region? Hint: the edit achieved two things as is explained and depicted in Figure 1, what were these two things?

2. For any gene-editing to be useful, especially for use as a therapy, it needs to be precise (i.e. it should edit the intended region, not some other region of the gene). Summarize what steps the authors took to determine how precise their editing was. 

3. How did the authors assess the efficacy with which the desired region was edited? (hint: they used at least 2 different ways to make this determination).

4. After creating the keratinocytes with the intended gene edits, what steps were taken to determine if the cells could actually fulfil the intended function when grafted on to the mice? If translated to the clinic, this procedure would involve obtaining patient keratinocytes and grafting the edited keratinocytes back on to the patients skin. What are some features of the process described in the paper that make it favorable for potential use on patients? What are aspects of the process that may still need further studies before being used on patients?
