  I  agree  that Gregory's transformation highlights his isolation and  alienation before his metamorphosis because is how he felt with the high  responsibility he had on his shoulder of being the person who had to  support the entire family, without having a personal or social life.  Having a hard worker life with a job that was exhausting waking up every  day very early in the morning , with those exhausted train trips. He  also had to endure the  mistreatment of his boss by telling him that  ultimately, he was not doing very well. He barely was eating well, and  more  just to support his family. 
Gregory did not have that  confidence that if one day he could not do it, he would feel released  that someone in his family would tap his shoulder by telling him now  rest you have been doing too much. He was doing more like a father of a  family than a simple son. The story reflects that he felt as an insect  and that was exactly happened after his transformation everyone  got  good jobs, instead of everyone having worked and helping each other as a  true family, he became the insect that brought money home with his  work, thus dying the love for a family member.
