Intelligent Packing Chromatography media is packed in the column using axial compression achieved by the column servo motor. Packing can be controlled using the appropriate AxiChrom Master wizard functions. Packing can also be performed and controlled using the UNICORN Intelligent Packing function of an ÄKTAprocess system for column sizes 300 to 1000.

ÄKTAprocess UNICORN Intelligent Packing functions are currently not com- patible for use with AxiChrom 1200-1600 columns as standard. Contact your local GE representative for more information about this.

Moving 1200-1600 columns After unpacking from the transport crate, AxiChrom 1200-1600 columns can be moved to the intended process position by lifting using eyebolts and an overhead crane or hoist. Alternatively AxiChrom 1200-1600 columns can be rolled over an even surface using a wheel kit that is available as an optional accessory for the column, see Wheel kits for AxiChrom 1200-1600 columns, on page 63, or the columns can be moved using other suitable capacity moving equipment, such as air castors.

NOTICE Do not place any type of lifting or moving equipment under the column feet.

Please note that the columns are heavy and cannot be easilymoved. Carefully read the Site Preparation Guide and Unpacking Instructions before placing the column.

6.2 General maintenance tools This section describes general tools required for the maintenance of all sizes of AxiChrom columns.

Tool kit General tools required for disassembly, maintenance and assembly of AxiChrom columns are provided in an AxiChrom toolkit, available from GE. For further details, see Section 12.7 Ordering information, on page 328.

O-ring removal tool The removal tool is used for removing O-rings from their grooves and for similar tasks. The tool is made of plastic to avoid scratching O-ring fittings. The image below shows how to use the removal tool to remove the snap ring used with plastic bed supports.

Plastic mallet The plastic mallet is used to tap the snap ring gently into position when fitting plastic bed supports. The groove on the mallet head may be used to lever the snap ring over the edge of the bed support if the fit is tight. The image below shows how to use the mallet head to lever the snap ring into position.

PTFE tape It is recommended to wrap PTFE tape around the threads of all bolts and screws of size M8 and above, with the exception of bolts that go into a threaded insert.

In maintenance procedures that involve the removal of bolts and screws with PTFE tape, remove the old tape from the bolts and apply new tape before refitting the bolts and attaching the nuts. Apply the tape in the same direction as the threads so that refitting the nuts draws the tape into the threads.

Column disassembly wizard

TheMaintenancemenu wizard dialog screens that the user sees are slightly different for AxiChrom300-1000 columns andAxiChrom1200-1600 columns. This is due to the extramaintenancewizard steps that allow the user to perform media valve maintenance on AxiChrom 1200-1600 columns.

NOTICE Be careful not to damage the column tube when re- moving the nuts. Use the closed end of the wrench to remove the nuts.

Verify that all top flange nuts are removed. Press Confirm to move the adapter to the top position.

Remove the lock blocks from their storage positions. Fit the lock blocks in the lock positions as illustrated below (the illustration shows four blocks in lock positions 1A, 1B, 1C and 1D: the number of blocks differs between col- umn sizes). Tighten the bolts securely to 45 Nm, using a torque wrench. Press Confirm.

NOTICE Do not use force to fit lock blocks. Lock blocks that are difficult to screw into place may be damaged and must be replaced.

Verify that the lock blocks are securely fitted. Press Confirm to start lifting the top unit assembly.

Result: The top unit now moves upward.

NOTICE Lifting the top unit with incorrectly placed lock blocks may damage the column.

Insert and secure the locking pins in the lock hole positions 2A and 2B, as illustrated below.

NOTICE Moving the adapter with only one locking pin in place may damage the column.

If the locking pins cannot be inserted into the lock holes for 300-600 columns, the top unit may not be properly seated on the lock blocks as shown below.

Shake the top unit gently by hand until the adapter rests securely on the lock blocks, as shown below.

Verify that the locking pins are correctly inserted and secured in position. Press Confirm to move the adapter up to the chamfer, in the top of the col- umn tube, to release the compression of the adapter seals.

Result: The adapter moves up to the column tube chamfer.

Unscrew the nuts and remove the bottom flange bolts, using two wrenches (one on either side of the bolt and nut assembly). Press Confirm.

Verify that all bottom flange bolts are removed. Press Confirm to lift the column tube.

Result: The column tube now moves up.

NOTICE If all bottom flange bolts are not removed, the column may be damaged in later steps.

NOTICE Make sure to insert the safety peg into the locking pin so that the safety catch is at the bottom of the locking pin, as shown in the illustrations. Insertion in any other orientation may result in damage to the safety peg and locking pin.

Swing out the column tube to perform the maintenance operations. Use the remaining locking pin to secure the column tube in the maintenance position.

CAUTION Make sure that no body parts become trapped be- tween the column tube assembly and the column frame when using the swing out function for mainte- nance work on the column. The column tube must be locked in place during all maintenance work. Due to the weight of the column tube assembly, it is not advis- able to try to stop the momentum of the assembly by hand when it is set in motion.

If the locking pins are difficult to remove, the top unit may not be properly seated on the lock blocks. If this happens for smaller columns with compara- tively lightweight top units (300-600 column sizes only), shake the top unit gently by hand until the adapter rests securely on the lock blocks, as shown in the image.

Verify that the locking pins are removed from both positions and placed in their storage positions. Press Confirm to lower the top unit.

Hold the media valve in position while removing the four bolts screwed into the valve from underneath the column (E).

Note:

Be aware that the residual liquid remaining in the media valve will spill out when the bolts are removed.

Continue to hold the media valve in place while loosening the two locking screws on the tube holder bolts (F).

The media valve and tube holder can now be removed downwards from the column. Let the tube holder slide off its fastener. The following steps on the media valve can be performed on a table.

Firmly press the release washer and hold it into the connector while removing the pneumatic tube from pneumatic cylinder (1). Repeat the procedure to disconnect the other pneumatic tube (2).

Clean the O-rings with 20% ethanol. Replace with new O-rings if necessary.

For 300-600 columns equipped with plastic tubes, the rinse inlet has an inner connector and additional O-ring.

Unscrew the four bolts to remove the pneumatic cylinder and its piston from the media valve outer body.

On AxiChrom 300-600 columns: When removing the piston, hold the upper part of the pneumatic cylinder, to prevent the cylinder from falling apart.

Loosen the four bolts on the bottom of the media valve outer body. Do not remove the bolts. Make sure a few millimeters of the bolt remain screwed into the media valve. Press the bolts against a flat surface so that the bolts push out the valve inner body, then remove the valve inner body.

Assembling the media valve At least two persons are needed when assembling the media valve.Note:

Refit all O-rings and the O-ring holder on the valve inner body.1

When refitting the diagonal O-ring on the valve inner body, make sure to stretch it when putting it back in place. It must not be rolled into place.

Refit the pneumatic cylinder into the media valve outer body.4

Make sure that the alignment of the pneumatic cylinder corresponds to the image below. Tighten the bolts (crosswise) in steps to 1.7 Nm.

Reposition the slurry inlet, rinse inlet and bottom mobile phase inlet/outlet tubes on the media valve. Tighten the bolts enough to hold the tubes firmly in position, but do not tighten the bolts fully at this stage.

CAUTION The air from damaged pneumatic tubes may cause injuries. Replace any damaged tubes.

Place the media valve so that the mobile phase tube rests in the correct position in the tube holder (A). Refit the tube holder bracket (B) on the mobile phase tube and tighten the bolts slightly, so that the tube can still slide under the bracket, to allow for adjustments in position when the tube holder is re- fitted to the bottom backing plate.

Reposition the Slurry inlet tube (A) and Rinse inlet tube (C) against the media valve body and tighten the tube bolts enough to hold the tubes firmly in position, but do not tighten the bolts fully until verifying that the tubes have been refitted at their correct angles.

Note:

To ensure that the Slurry inlet tube and the Rinse inlet tube are placed in their correct angle when refitted, assemble the tube holder brackets, but tighten the bolts only slightly, so that the tubes can still slide under the brackets.

When all of the media valve tubes are correctly in position, tighten all of the bolts holding the tubes to the media valve to 2.2 Nm.

For AxiChrom 1200-1600 columns: It is recommended to perform a media valve leakage test at this stage, using the leakage test unit that is available for this purpose (optional accessory).

Refit the two pneumatic tubes on the pneumatic cylinder according to the image below. Make sure that the two tubes are connected to the correct connectors.

1 Assemble the four media valve bolts into the bottom backing plate. Tighten the bolts (crosswise) in steps up to 5 Nm.

2 Tighten the bolts on the tube holder brackets. The bolts should be tight- ened evenly and not too hard, to avoid any damage to the tubes, espe- cially if plastic tubes are used.

3 For media valves equipped with plastic tubes, make sure the tubes are at a slight angle to the horizontal so that liquid in the tubes drains away from the media valve.

4 Tightening the locking screws on the tube holder bolts to 10 Nm.

Connect the air supply tubes to the two valve pneumatic inlets on the tube holder. Open the air supply to the column.

Connect the hoses to the Slurry inlet, Bottom mobile phase inlet/outlet , and Rinse inlet. See Section 4.6 Connections, on page 68 for more information.

Open the Luer connector on the tube of one of the flushing ports (for example, flushing port A in the illustration of the locations of the flushing ports on AxiChrom 1200-1600 columns).

6.7 Changing flushing connector O-rings There are two flushing connectors for the 300-1000 columns and four flushing connectors for the 1200-1600 columns, on the top of the adapter backing plate. Inside each flushing connector are two O-rings, which should be changed at least once every two years. The O-rings should be checked regularly and cleaned if needed.

Follow the instruction below to change the flushing connector O-rings.

Tip:

Leave a couple of the nuts on opposite sides partly unscrewed. This will de- crease the risk that the bed support assembly falls down by accident.

With the weight supported and at the same time gently pressing on the partly unscrewed nuts might help to remove the bed support assembly if stuck.

WARNING The bed support and distributor assemblies are heavy, particularly on larger columns. Make sure that these components are securely supported and cannot fall when fastening nuts or bolts are removed.

Note:

The nuts should release from the fasteners, leaving the fasteners attached to the bed support. If any fastener releases from the bed support, refit the fas- tener and tighten to a torque of 20 Nm.

Then use a torque wrench to tighten the M12 bolts on the distributor plate to 15 Nm and the four bolts underneath the inlet/outlet assembly to 11 Nm.

Refit the plastic spacer and O-ring (if used) on the inlet/outlet. The plastic spacer is used on the 300, 400, 450 and 800 columns, and not on the 600 and 1000 columns. In the image below, arrows indicate the plastic spacer and O-ring.

Refit the adapter inlet/outlet to the column. Make sure that the O-rings are positioned correctly. Refit the inlet/outlet so that the tubing is closest to the tube roller assembly. Tighten the four bolts crosswise to 5 Nm.

NOTICE When fitting an old inlet/outlet on a newer column, check if there are more than four threaded holes that the inlet/outlet can be fitted to on top of the adapter. If so, remove the guide pin from the inlet/outlet if it exists.

Refitting the bed support

Refit the two center O-rings, one on each side of the bed support.1

Refit the bed support O-ring and scraper seal.2

While holding the bed support in place, refit the bed support fastening nuts, and their associated plastic and stainless steel washers, so that the bed support is retained. Do not tighten the nuts yet.

Wet the threads of the bed support tool, and then use the threaded side of the bed support tool to center the bed support on the distributor plate. Make sure that the center of the bed support exactly fits into the corresponding countersink in the distributor plate, then tighten the bed support fastening nuts fingertight, while the bed support tool is still in place.

Tighten the bed support fastening nuts using a torque wrench to tighten all of these nuts crosswise to a torque of 10 Nm for 300-600 columns, or 15 Nm for 800 and 1000 columns.

8.1 Maintenance tools for 1200-1600 columns

Introduction This section describes tools that are used specifically when performing service and maintenance on AxiChrom 1200-1600 columns. General tools used with all column sizes are described in Section 6.2 General maintenance tools, on page 111.

NOTICE Always use appropriate tools and the correct tightening torques for bolts when working with the AxiChrom column and AxiChrom Master unit. Only the special toolkit provided by GE for the AxiChrom column should be used.

Bed support tool The bed support tool is used for removing and replacing the bed support screw. Engage the pins on the tool firmly in the screw when using the tool.

The bed support tool for 1200-1600 columns is shown below.

The threaded end of the tool is used with stainless steel bed supports to assist in centering the bed support on the distributor during assembly.

Bed support nut tool The bed support nut tool is used for tightening the bed support nuts during assembly and disassembly of the plastic bed supports on 1200-1600 columns.

Bed support lifting device The bed support lifting device is used for the disassembly and assembly of AxiChrom 1200-1600 columns.

The bed support lifting device is used for example during:

• removal/fitting of plastic and stainless steel adapter bed supports, and the adapter distributor plate,

• removal/fitting of stainless steel bottom bed supports, and the bottom distributor plate.

The maximum lifting capacity of the bed support lifting device is 120 kg.

If a bed support lifting device is not used when performing the disassembly and assembly of AxiChrom 1200-1600 columns, it is recommended that a minimumof five people are on hand towork together to performmanual lifting operations. As it is strongly recommended that a lifting device is used to per- form maintenance steps, manual lifting operations are not described.

Bed support protection

A bed support protection mat and disposable protective is provided with the column.

These are also recommended accessories to keep on site for use during the maintenance of AxiChrom 1200-1600 columns to protect the bottom bed support.

The bed protection consists of two layers:

• a thin disposable plastic film (sold in sets of five) that is placed directly onto the bed support

• a thicker reusable mat that is placed on top of the plastic film.

The mats can be ordered from GE for the appropriate column size.

Nozzle O-ring assembly tool The replacement of rinse nozzle tip O-rings is performed by manually removing the O- rings from the respective nozzle tip grooves, cleaning the grooves and fitting new O- rings.

A conical nozzle O-ring expansion tool is available for expanding and fitting the new O- rings into place on the nozzle tip during rinse nozzle assembly, see Assembling the rinse nozzle, on page 212.

To switch between nozzle tube positions the tube holder is simply unscrewed and rotated 180 degrees. The following image shows the lower tube position (A), used when columns are fitted with plastic bed supports. • bottom mobile phase inlet/outlet,

NOTICE If the column is equipped with a bypass valve, make sure that the position of the bypass valve will not re- strict the movement of the bottom backing plate.

Wheel the bed support lifting device into position, a few centimeters above the bed support.

Note:

To correctly align the tri-arm screw fittings of the bed support lifting device with the corresponding threaded holes in the bed support or distributor plate of the bottom columnassembly, the bed support lifting devicemust bemoved into position from the side of the column as shown in the following illustration.

Align the center of the tri-arm with the center of the bottom bed support.10

Check that the tri-arm threaded screws are aligned with their corresponding threaded holes (M10) in the bed support, but make sure that the tri-arm screws are retracted so that it is the center fitting that first comes into con- tact with the bed support when the bed support lifting device is lowered.

Lower the bed support lifting device tri-arm, using the rear winch on the lifting device, until the bed support fitting on the lifting device enters and rests against the center of the bed support and the tri-arms are levelly posi- tioned above the bed support.

The bed support fastening nuts should release from the fasteners, leaving the fasteners attached to the distributor plate. If any fastener releases from the bed support ring instead, refit the fastener and tighten to a torque of 20 Nm.

Adjust the tri-arm lower clamp fittings until all three clamps sit tightly against the distributor, with the tri-arms of the bed support lifting device level with the distributor plate.

Unscrew the bolts to remove the distributor plate. The number of bolts varies with column size.

WARNING The bed support and distributor assemblies are heavy, particularly on larger columns. Make sure that these components are securely supported and cannot fall when fastening nuts or bolts are removed.

Tighten the stabilizing screws in the center of the tri-arms of the bed support lifting device.

Slowly lower the tri-arm a few centimeters, while making sure the distributor plate separates from the adapter.

Lower the distributor until it is clear of the column, and then remove the scraper seal and O-ring from the distributor plate.

Clean the scraper seal, the large O-ring around the distributor plate and the O-ring groove with 20% ethanol.

Screw the upper clamps onto the tri-arm fittings, to clamp the distributor plate securely in place on the bed support lifting device, and then pull out the bed support lifting device slowly, taking care not to bump the distributor plate into the column stand.

Move the lifting device away from the column, and then rotate the tri-arms of the lifting device through 180 degrees so that the distributor plate can be carefully maneuvered onto a clean, protected surface and removed from the bed support lifting device.

Make sure that the clean distributor plate is securely fastened to the bed support lifting device using the upper tri-arm clamps and the distributor fit- ting. Rotate the tri-arm of the lifting device if required, so that the distributor plate is in the correct orientation for fitting to the adapter.

Release the distributor plate from the tri-arms of the lifting device using the spring loaded pin on the center fitting and lower the tri-arm a few centimeters using the rear side winch. Remove the distributor fitting from the center of the distributor plate, then carefully move the bed support lifting device away from the column.

To make installation easier, make sure that the thin O-ring is thoroughly wetted with 20% ethanol during assembly. The thin O-ring may pop off if it is dry during assembly.

Attach the plastic and stainless steel washers and fastening nuts to the threads of the bed support fasteners, with the plastic washers closest to the backing plate. Finger-tighten the nuts onto the fasteners. Make sure that there is no gap between the assembly and the backing plate all the way around the circumference of the column.

Pull out the spring loaded pin on the distributor fitting, and then lower the bed support lifting device until it is well clear of the adapter, using the rear side winch. Carefully move the bed support lifting device away from the column.

Wet the threads of the bed support tool, and then use the reverse end of the bed support tool to center the bed support. With the bed support tool in place, use a torque wrench to tighten all of the bed support fastening nuts crosswise, in increments to a torque of 30 Nm, then remove the bed support tool.

NOTICE Make sure that the bed support is raised evenly against the distributor as the bed support fastening nuts are tightened. Perform a final check around the entire cir- cumference of the column after the final tension has been reached to make sure that there are no gaps and that the bed support is fully in contact with the distrib- utor around the entire column.

Refit the bed support

NOTICE Clean O-ring grooves and lubricate all O-rings thoroughly with 20% ethanol before fitting O-rings into place.

ActionStep

Lubricate and then refit the O-ring on the distributor plate.1

Lubricate and refit the two small O-rings into their respective O-ring groove in the center of the the bed support, then lubricate and fit the large bed support O-ring into the bed support O-ring groove. Make sure that the large O-ring is properly seated in its groove.

Note:

There are two central bed support O-rings, one on each side of the center of the bed support. Both of these O-rings cannot be inserted at the same time when the distributor plate is attached to the lifting device. The O-ring that is situated on the lifting device side of the distributor plate can be attached before or after the lifting device is used.

Loosen the counter nuts of the tri-arm threaded screws, and unscrew the tri-arm threaded screw fastenings from the bed support.

Tighten the counter nuts again and make sure that the tri-arm threaded screw fittings are securely attached to the tri-arms again and are clear of the bed support.

Use the rear side winch to lift the tri-arm until it is well clear of the bed sup- port. Carefully move the bed support lifting device away from the column.

Check that the bed support is still flat against the distributor plate, then fit the washers and bed support fastening nuts to the bed support fasteners protruding from the underside of the backing plate. Finger tighten the fas- tening nuts.

Insert the threaded end of the bed support tool into the center of the backing plate and screw into place.

With the bed support tool in place, use a torque wrench to tighten all of the bed support fastening nuts crosswise, in increments to a torque of 15 Nm.

Note:

The bed support fastening nuts can loosen when the tube is fastened into place. Check torque tensions and retighten to 15 Nm as required.

Unscrew and remove the bed support tool from the center of the bed support.14

Lubricate the bed support screw O-ring and the bed support screw with 20% ethanol, and then fit the O-ring onto the bed support screw.

Make sure the media valve is open. The media valve is open when the piston in the center of the bed support is lowered. If the media valve is closed, it can be opened in the MANUAL CONTROL wizard in AxiChrom Master.

Remove the bed support by hand, lifting it straight upwards and making sure that any remaining screws do not damage the bed support. If necessary, insert the O-ring removal tool between the edge of the bed support and the O-ring and gently lever the bed support out of its seating.

Remove the inner and outer O-rings from the bed support screw. Clean the O-rings and the bed support screw with 20% ethanol. Replace with new O- rings if necessary.

Refit the four bolts into the bolt positions underneath the adapter rinse nozzle and finger tighten these bolts, making sure that the distributor is correctly positioned and centered.

Pull out the spring loaded pin on the distributor fitting and lower the lifting device using the rear side winch, and then slowly remove the bed support lifting device.

Refit the bolts on top of the distributor plate. Fit both stainless steel and plastic washers on each screw, with the plastic washer closest to the distrib- utor plate. Tighten the bolts to 15 Nm.

Tighten all of the bed support fastening nuts, crosswise, to a torque of 6 Nm.9

Remove the distributor fitting from the distributor plate.10

Tighten the four central bolts, the bolt positions underneath the adapter rinse nozzle, to a torque of 11 Nm.

Refit the four flushing connectors according to the assembly instructions in Section 6.7 Changing flushing connector O-rings, on page 158.

Refit the O-ring on the bed support screw, then wet the O-ring and screw with 20% ethanol.

Spray the surfaces at the center of the bed support with 20% ethanol, then refit the bed support screw into the center of the bed support. Carefully start to tighten the bed support screw by hand making sure that the threads are not crossed.

Tighten the bed support screw firmly using the bed support tool. Make sure the bed support is secured by the bed support screw.

NOTICE Be careful not to damage the bed support or the bed support screw when using the bed support tool.

Refit the bed support nuts (replace where necessary). The bed support nuts should be inserted so that they all just enter the extrusion in the sinter. Do not tighten the nuts.

Wet the bed support screw, and then refit the bed support screw. Carefully start to tighten the bed support screw by hand, making sure that the threads match. Finally tighten the bed support screw until it reaches a firm stop.

Tighten the bed support nuts to a torque of 1.8 Nm.6
